MGI Tech Co., Ltd., a global developer of life science technologies, has launched two new nanopore sequencing products, CycloneSEQ-WT02 and CycloneSEQ-WY01, equipped with advanced CycloneSEQ technology. This new technology integrates enhanced protein engineering, a novel flow cell design, and an optimized basecalling algorithm to provide high accuracy and throughput for genomic applications.

Key Technological Advancements

The CycloneSEQ platform incorporates several notable improvements:

  • Enhanced Protein Performance: Utilizing a motor protein with high thermal stability and a structurally rigid nanopore protein, the platform excels in unwinding nucleic acids and maintaining low sequencing noise. This structural refinement enhances nucleic acid capture and threading efficiency, boosting overall sequencing reliability.

  • Improved Flow Cell Design: Each flow cell supports up to 4,096 nanopore protein sensors arranged in a high-density array, designed for ultra-long sequencing durations. The integration of BioMEMS and ASIC technologies ensures precise detection of currents at picoampere levels, facilitating stable, real-time sequencing.

  • Optimized Basecalling Algorithm: The platform employs a proprietary basecalling algorithm, developed through extensive data and large-scale distributed training, achieving a 97 percent accuracy rate and enabling real-time decoding across multiple flow cells.

  • Innovative Chemistry: A new sequencing chemistry, termed nanopore local chemistry, modulates electrophoretic migration of magnesium ions to control motor protein activity, further enhancing sequencing efficiency.

Applications and Performance

The CycloneSEQ-WT02 and CycloneSEQ-WY01 sequencers are designed for a wide range of genomic applications, including public health, cancer research, reproductive health, and environmental studies. The CycloneSEQ-WT02 features a dual flow cell architecture for simultaneous operations, enabling fast coverage and real-time analysis suitable for complex sequence detection and de novo genome assembly. The CycloneSEQ-WY01, characterized by a high-density flow cell, supports ultra-long read lengths and high-throughput sequencing.

Both models offer sequencing capabilities with read lengths from base pairs to megabase pairs and operational speeds between 350 and 420 nucleotides per second. The CycloneSEQ-WT02 provides up to 50 gigabases throughput per single flow cell, while the CycloneSEQ-WY01 achieves even higher throughput with over 30,000 nanopores per flow cell.

The CycloneSEQ-WT02 will be available for order in select countries later this year, and the CycloneSEQ-WY01 is expected to launch in the first half of 2025. These additions further expand MGI's sequencing technology portfolio, reinforcing its commitment to providing comprehensive multi-omics solutions globally.
